Absolute hand determination of glycofibrils from natural sources in cryo-em

This study proposed the Ahaha method for determining the absolute hand of naturally sourced glycofibrils in cryo-EM. Existing methods (such as tilt-pair imaging) are difficult to apply to heterogeneous samples. Ahaha utilizes mono-tilt micrograph data, based on the prior knowledge that fibrils tend to lie flat in the ice layer, and determines the absolute hand by examining the consistency between particle tilt and stage tilt. The study validated the method's correctness using a karst cave water sample containing both bacterial pili (whose hand could be verified by alpha-helix handedness) and glycofibrils, and determined the absolute hand of four glycofibrils (TLP-2a, TLP-2f, TLP-2g, TLP-2h), constructing their atomic models. These glycofibrils feature a core linear peptide chain composed of dipeptide repeats, modified by O-glycosylation, revealing a potential family of glycofibrils. The Structural Biology Core Facility provided critical equipment and technical support: two Krios G4 microscopes were used for data acquisition; platform engineers assisted with image acquisition.
