Platform Introduction

Flow Cytometry Core Faclity provides multi-parameter and multi-dimensional cell marker-based cell analysis and sorting services. Integrating single-cell sample preparation, multicolor immunofluorescent labeling, high-throughput flow cytometry analysis, and targeted cell sorting technologies, the platform supports basic research and clinical applications in immunology, oncology, neurobiology and other fields.

To meet diverse demands for biological sample analysis and sorting, the platform has established a complete flow cytometry equipment system, covering five categories: conventional flow cytometry, spectral flow cytometry, imaging flow cytometry, nano-flow cytometry and microbial flow cytometry. With clear division of labor and complementary advantages, these instruments fully cover core experimental scenarios in life science research, delivering efficient and precise technical support for scientific research.


1) Conventional Flow Cytometry Analyzer and Sorter

This category mainly includes analysis and sorting modules. Conventional analyzers include BD FACSymphony A1 and FACS LSRFortessa X-20, enabling rapid multi-parameter phenotypic identification of various samples. Conventional sorters include BD FACSAria Fusion and BD FACSAria III, which can efficiently achieve precise sorting and purification of target samples. As core instruments for sample processing and phenotypic analysis in basic scientific research, they are suitable for most routine experimental requirements.


2) Spectral Flow Cytometry Analyzer and Sorter

As an upgraded supplement to conventional flow cytometry, it effectively breaks through the bottlenecks of traditional flow cytometry detection. Spectral analyzers include Cytek Aurora and BD FACSymphony A5 SE, supporting in-depth multi-parameter analysis of complex samples and reducing spectral overlap interference. Spectral sorters include BD FACSDiscover S8 and Cytek Aurora CS, balancing analytical precision and sorting efficiency, and are applicable to in-depth research scenarios of highly complex samples.


3) Imaging Flow Cytometry Analyzer and Sorter

Its core feature is the integration of high-throughput flow analysis and high-resolution imaging, overcoming the limitation of “detection without visualization” in conventional flow cytometry. The imaging analyzer is Cytek ImageStream MKII, which can simultaneously acquire up to 10 fluorescence parameters and high-resolution bright-field and fluorescence images of single cells. It not only achieves accurate cell phenotypic analysis, but also directly visualizes cell morphology, subcellular structures and marker localization, realizing synchronous “qualitative + quantitative + localization” detection. The imaging sorter is BD FACSDiscover S8, which integrates imaging verification on the basis of retaining high-dimensional precise analysis and sorting capabilities, and realizing the integrated function of “analysis + imaging + sorting”. It can confirm the morphological characteristics of target cells in real time during sorting, effectively avoiding mis-sorting and greatly improving the purity of sorted samples and the reliability of experimental results. It is especially suitable for precise analysis and sorting of rare cells, cell-cell interactions, morphologically heterogeneous cells and protein phase transition.


4) Flow NanoAnalyzer

The platform is exclusively equipped with a Nanoparticle Cell Analyzer U30E Pro dedicated to the precise characterization of nanoscale particles. It allows for accurate multi-parameter detection of particle size, concentration and surface biomarkers, supporting research in exosomes, vesicles, nanobiology and materials science, and filling the equipment gap in nanoscale sample analysis for the platform.


5) Microbial Sorter

Equipped with Precision Cell Sorter Sony MA900, it is specially designed for microbial samples and can efficiently perform microbial sorting, enrichment and identification experiments. It provides dedicated technical support for microbiological research and environmental microbial detection, improving the efficiency and accuracy of microbial experiments.

The platform’s flow cytometry equipment system covers a wide range, fully meeting core scientific research needs including cell phenotypic identification and sorting, high-dimensional analysis and sorting of rare cells, nanoparticle characterization and identification, imaging morphological analysis and sorting, and microbial analysis and sorting. It has developed 11 full-process technical capabilities, listed in Table 4.1.

In addition, the platform provides full-process training for various instruments year-round. Based on equipment classification and assessment of users’ proficiency, users are granted primary, intermediate and advanced operation qualifications respectively. Primary users operate instruments with on-site assistance, while intermediate and advanced users can operate independently.

In 2025, a total of 92 training sessions were held, covering 137 person-times of primary users, 94 person-times of intermediate users and 7 person-times of advanced users. To ensure standardized data processing, the platform regularly conducts specialized FlowJo data processing training to guide users in implementing proper data processing workflows.

Since its establishment, the platform has served 52 research groups and signed service agreements with 11 external research groups. Recently, four core technologies have been prioritized for development:

1. High-dimensional spectral flow cytometry deep immune phenotyping technology, enabling precise profiling of rare immune cell subsets in complex samples;

2. Imaging flow cytometry simultaneous cell morphology-phenotype detection technology, improving the accuracy of abnormal cell detection;

3. Precision nanoparticle characterization technology, realizing multi-dimensional accurate detection via dedicated instruments and adapting to multi-field research;

4. Efficient microbial sorting and identification technology, supporting microbial sorting, purification and population analysis for interdisciplinary research.

The platform will continue to provide standardized technical support to facilitate scientific research and technological transformation.

She earned her bachelor’s degree in Biochemical Engineering from the Department of Chemical Engineering, Tsinghua University, and obtained her Ph.D. in Biochemical Engineering from University of Connecticut. She completed her postdoctoral training at Albert Einstein College of Medicine.

Since 2010, she has successively served as Flow Cytometry Core Manager at Icahn School of Medicine at Mount Sinai (New York) and Technical Director of Stem Cell & Flow Cytometry Core at Albert Einstein College of Medicine (New York). In March 2026, she joined Shenzhen Medical Academy of Research and Translation full-time as a member of the Flow Cytometry Core Facility at the Bio-Tech Center.

Her research focuses on the system architecture of complex flow cytometry platforms and the development of high-dimensional flow cytometry technologies, with key emphasis on the establishment of platform systematic standardization, breakthroughs in high-dimensional cell sorting techniques, design of high-dimensional color panels, and translation of cutting-edge technologies.

She has published numerous papers in internationally renowned journals including Cell Stem Cell, Nature Medicine, and Cancer Cell, et al. She also holds the Specialist Certification in Cytometry jointly awarded by the International Society for the

Advancement of Cytometry (ISAC) and the American Society for Clinical Pathology (ASCP).

Dr. Liu Jie received her Ph.D. in Biomedical Sciences from the Faculty of Health Sciences, University of Macau. Her doctoral research focused primarily on CAR-T cell therapy.

She joined the Flow Cytometry Core Facility in 2024, where she mainly oversees the daily operation and maintenance of flow cytometry instruments, including BD FACSAria Fusion and BD FACSAria III cell sorters, Cytek Aurora and BD FACSymphony A5 SE spectral flow cytometers, and Cytek ImageStream MKII imaging flow cytometer.

Dr. Liu has extensive practical experience in flow cytometry sample preparation, panel design, and multicolor immunofluorescence analysis and cell sorting. She is currently leading a Youth Program of the National Natural Science Foundation of China (NSFC). She has published more than 10 peer-reviewed SCI papers in renowned journals including Journal of Hematology & Oncology, Small, MedComm, Chemical Communications, and Frontiers in Immunology.
